Did you put a new wire on? What did the plug on 3 look like, normal or different than the rest?

3 guesses would be plug wire, head gasket (or head) problem, or valvetrain. How many miles on the engine and which engine?

I'd do a compression or leak down test, and check the oil and coolant levels, not driving it until fixed if you find coolant in the oil or vice versa, though if it goes on too long a misfire can damage a catalytic converter too. In rarer cases you might have a head leak where exhaust goes out through the cooling system and is seen as bubbles in the radiator or with a chemical test.

If the problem isn't found the typical thing to do is start tearing down the engine looking for it, in the vehicle if possible, take the intake manifold and head off.
Please post your vehicle year/model/engine/(n)WD in help topics.
